Cell Biolabs Inc Glycogen Assay Kit (Fluorometric), 100 assays
Cell Biolabs’ Glycogen Assay Kit (Fluorometric) is a simple assay that measures the amount of total glycogen present in biological samples in a 96-well fluorescence plate reader. Glycogen is broken down into glucose monomers by amyloglucosidase first, glucose is then oxidized by glucose oxidase into D-gluconic acid and hydrogen peroxide. The resulting hydrogen peroxide is then detected with a highly specific fluorometric probe in a standard 96-well colorimetric plate reader. Sample glycogen concentrations are determined by comparison with a known glycogen standard. The kit has a detection sensitivity limit of 120 nM glycogen. Each kit provides sufficient reagents to perform up to 100 assays, including blanks, glycogen standards, and unknown samples. Note each sample replicate requires 2 assays, one of which is a negative control to be subtracted as background.

In the floating matrix contraction model, a freshly polymerized collagen matrix containing cells is released from the culture dish and allowed to float in culture medium, and contraction occurs in the absence of external mechanical load and without appearance of stress fibers in the cells. The CytoSelect™ 24-Well Cell Contraction Assay Kit provides a simple system to assess cell contractivity in vitro and screen cell contraction mediators using the floating matrix model. The kit's proprietary Cell Contraction Plate eliminates the matrix releasing step of the conventional contraction assay, providing a faster, higher-throughput method to assess cell contraction. Cayman’s HDAC Cell-Based Assay Kit provides a robust and EAsy-to-use platform for studying HDAC activity modulators in whole cells. By using a cell-permeable HDAC substrate, the activity of various lysine-specific protein deacetylases can be measured in intact cells in a simple and homogenous manner. The fluorescence of the deacetylated reaction product can be analyzed using a fluorometer with an excitation wavelength of 340-360 nm and an emission wavelength of 440-460 nm. An HDAC inhibitor, trichostatin A, is included for checking specificity of the HDAC reaction. This assay parallels Cayman’s HDAC Activity Assay Kit (Item No. 10011563), which uses a nuclear extract rather than whole cells for the assay. Together, both assays will help identify potential inhibitors/activators of HDAC enzymes.
